systems, has announced a new technical paper describing a novel
flow cytometric type assay that provides rapid, accurate determination
of Baculovirus titres.

Recombinant
protein expression using the Baculovirus vector system is a powerful
tool for the production of therapeutics, diagnostics and reagents.
To maximise efficiency of protein production, and thereby reduce
costs, optimal manufacturing parameters are typically determined
using the Baculovirus titre assay.
The
technical paper discusses the benefits and shortfalls of traditional
Baculovirus titre methods including real-time PCR, end point dilution
and plaque assays. Results obtainable by these methods are compared
to the flow cytometric type assay carried out using a Guava PCA-96
system. The Guava PCA-96 is an affordable, ultra-compact system
ideally suited for automatic, 96-well microplate and multiple microcentrifuge
tube analysis of cell lines and compounds. Intuitive in operation,
the Guava PCA-96 provides precise, accurate and easy-to-read results
with often less than one day of training. The Guava PCA-96 system
simplifies cell monitoring and screening, only a few microlitres
of samples are required for each experiment, saving precious cells
and reagents and reducing potentially hazardous biological waste.
Using
a selection of Spodoptera frugiperda and Trichoplusia insect cell
lines the flow cytometric type assay, performed on the Guava PCA-96,
was found to give results of equal or greater accuracy than the
traditional methods. The independently researched technical paper
concludes that the high-throughput Baculovirus titre, using the
Guava PCA-96, facilitated accurate virus titre determination of
multiple conditions in less than 24 hours, far less than the four
days required for traditonal plaque assays. Further, not only did
the new assay enable determination
of
infectious particles versus viral transcript, the high-throughput
format also allowed for direct comparison of various titre specific
to target cells as well as media consumption.
